It intends to guide the development of methods, the refinement of old ones, and the adaptation in general of methods to almost any scale of sensitivity. This manual is organized into three parts: a general section, one on quantitative histochemistry, and an appendix containing information that may be useful to have at the bench. The general section is comprised of nine chapters that focus on properties of the pyridine nucleotides, kinetics, and glassware, as well as on improvement, modification, adaptation, trouble shooting, and development of methods. This part also describes the preparation of tissues for analysis, the enzymatic cycling methods, and a compendium of 36 metabolite assays. The quantitative histochemistry section is comprised of four chapters that include information on the preparation of frozen-dried material and dissection of samples for analysis; the fishpole balance for weighing samples; and the generalities of analysis with emphasis on the ""oil well technique."" This book will be useful to novices as well as experts who are familiar with other analytical styles.

It is intended as a guide to the development of new methods, to the refinement of old ones, and to the adaptation in general of methods to almost any scale of sensitivity. As some may realize, the book is a sequel to A Flexible System of Enzymatic Analysis, originally published in 1972. The major changes, other than an appropriate interchange of authors, consist of a wholly new chapter of methods and protocols for measuring enzymes, the addition of 13 new entries in the metabolite chapter, and a much superior chapter on enzymatic cycling. With considerable nostalgia, we have switched from DPN and TPN to NAD and NADP nomenclature, which no doubt will make Otto Warburg turn over in his grave. The incentives for the methodology in this book came from the rigorous demands of quantitative histochemistry and cytochemistry. These demands are specificity, simplicity, flexibility, and, of course, sensitivity—all likewise desirable attributes of methods for other purposes. The specificity is provided by the use of enzyme methods. Simplicity is achieved by leading all reactions to a final pyridine nucleotide step.

This is the fifth symposium on Experimental and Clinical Hepatology to be convened by the organisers, and, in common with the previous four, reflects the importance placed upon continuing discussion by clinical investigators as well as basic physiological and pharmacological researchers. Previous meetings which were held at the Surgical Department of the Philipps University in Marburg were dedicated to problems of direct clinical application, such as haemodynamics of portal hypertension, surgical approach to hepatic tumours, liver regeneration and temporary hepatic support. Clinical investigation was a subject which frequently came under discussion and some of these investigations have since lead to direct improvements in clinical therapy and many patients have benefited as a result. This time, the convocation was called to the Medizinische Hochschule Hannover where remarkable advances in clinical liver trans plantation have been the most striking event in clinical hepatology. The concensus of the meeting was that while significant clinical progress has been made, a variety of basic problems remained unsolved which will require further investigation. Special attention has, therefore, been given to topics such as acute liver damage, particu larly within the transplanted liver, to chronic active hepatitis and its spontaneous prognosis, to isolated hepatocytes and their effect on functional support of the liver, to mechanisms of liver circulation which may be altered completely within the transplant and to liver perfusion which is still considered unsatisfactory for subsequent transplantation.

To my mind, the superoxide radical discovered by Linus Pauling more than 50 years ago is about to become a major issue in Ameri can medicine. The use of cultured cells in the clinical diagnosis of hereditary metabolic dis ease is a rapidly developing subject to which many different disciplines have brought their expertise and knowledge. A number of scientists who have in dividually contributed to the growth of the subject gave invited papers at the Fourteenth Symposium of the Society for the Study of Inborn Errors of Metabolism in the University of Edinburgh on 13-16th July, 1976. These papers form the basis of this monograph which brings together contributions from the basic sciences and from physicians concerned primarily with human disease. The cross-fertilization produced by this interdisciplinary communica tion was invaluable to those trying to understand and overcome diagnostic problems posed by hereditary metabolic disease. Cell culture methods and cell preservation techniques were described by D. G. Harnden and D. E. Pegg; Dr T. Elsdale outlined some of the factors which control in vitro cell growth and division. Cell culture methods and cryopreser vation techniques have allowed the wide distribution of biochemically abnor mal cells and their study over long periods of time. It is also evident that when a defect which produces severe metabolic disorder in man can be studied in the laboratory using isolated cell cultures a wide variety of investigative procedures can be focused on to the cellular defect without distress or discomfort to the patient or relatives.
