Molecular toxicology is on the way to setting a new standard in toxic risk assessment, with the powerful help of concepts and methods developed in the fields of molecular and cellular biology. The methods of molecular toxicology are endowed with qualities not always secured in more traditional approaches to toxicity assessment. These methods are easily standardized, reliable, reproducible and require only tiny amounts of the xenobiotic. They are much simpler, faster and less costly than current tests on animals and can be implemented with modest laboratory resources. This volume of Advances in Molecular Toxicology represents a collection of invited papers presented at the First European Workshop on Molecular Toxicology in Sophia Antipolis, France, The book covers 8 selected topics in molecular toxicology. The first two are devoted to ''physics'', dealing respectively with advanced instrumentation enabling location and quantification of xenobiotics in cells and organs with ultimate precision, and computer-assisted assessment of risk (QSAR). A comprehensive and up-to-date survey of molecular toxicology in the pharmaceutical industry is contributed by a pioneer in this field. The third section addresses the effect of xenobiotics on cell-cycle actors. Sections 4--6 deal with molecular toxicology in skin, kidney, nerve, liver and pancreatic cells. The last two sections describe methods to probe DNA damage and repair, and immunotoxicity.

The science of toxicology has progressed considerably since Molecular Toxicology was first published in 1997. New advances in biochemical and molecular biological experimental techniques have helped researchers understand the precise effects of toxins and foreign compounds on living things at the molecular, cellular, and organismal levels. Breakthrough research has recently been completed illuminating the human genome and the role of enzymes in toxic biochemical reaction mechanisms. Toxicology now covers drug metabolism and design, carcinogenesis, programmed cell death, and DNA repair, among other subjects. The second edition captures these and other advances, and broadens its scope to address the experimental science of toxicology. The first edition of Molecular Toxicology has become an indispensable resource for graduate students in molecular and biochemical toxicology courses, as well as academic researchers and industrial researchers in toxicology. Rigorously updated and revised, the new edition commands an unrivaled authority in the field of molecular toxicology.

Molecular Biological Markers for Toxicology and Risk Assessment provides an introduction to the exciting field of biomarkers and their use in toxicology and risk assessment. In recent years, new classes of molecular biomarkers capable of detecting early manifestations of ongoing chemical-induced cell injury and cell death have been developed as a result of advances in analytical chemistry, molecular biology, and computational modeling. The interplay between these emergent tools of science has resulted in new insights into initial mechanisms of chemical-induced toxicity and carcinogenicity. Molecular Biological Markers for Toxicology and Risk Assessment guides the reader through a broad range of molecular biological markers, including the "omic" biomarkers, and provides an examination of the various elements in the evolution of these modern tools. It then explores possible ways in which these markers may be applied to advance the field of chemical risk assessment. Since molecular biomarkers and related technologies are inherently complex, the book concludes with a section on risk communication in order that readers may appreciate both the strengths and limitations of molecular biological marker approaches to risk assessment practice. Introduces the use of molecular biomarkers to detect toxic effects of chemicals as early as possible Provides an accessible overview of this emerging, interdisciplinary field, to best inform decision making in chemical and pharmaceutical safety Includes a section on risk communication of these complex concepts, essential for effective risk assessment Provides new insights into the initial mechanisms of chemical-induced toxicity and carcinogenicity

Advances in Mathematical Chemistry and Applications highlights the recent progress in the emerging discipline of discrete mathematical chemistry. Editors Subhash C. Basak, Guillermo Restrepo, and Jose Luis Villaveces have brought together 27 chapters written by 68 internationally renowned experts in these two volumes. Each volume comprises a wise integration of mathematical and chemical concepts and covers numerous applications in the field of drug discovery, bioinformatics, chemoinformatics, computational biology, mathematical proteomics, and ecotoxicology. Volume 1 includes chapters on mathematical structural descriptors of molecules and biomolecules, applications of partially ordered sets (posets) in chemistry, optimal characterization of molecular complexity using graph theory, different connectivity matrices and their polynomials, use of 2D fingerprints in similarity-based virtual screening, mathematical approaches to molecular structure generation, comparability graphs, applications of molecular topology in drug design, density functional theory of chemical reactivity, application of mathematical descriptors in the quantification of drug-likeness, utility of pharmacophores in drug design, and much more. Brings together both the theoretical and practical aspects of the fundamental concepts of mathematical chemistry Covers applications in diverse areas of physics, chemistry, drug discovery, predictive toxicology, systems biology, chemoinformatics, and bioinformatics Revised 2015 edition includes a new chapter on the current landscape of hierarchical QSAR modelling About half of the book focuses primarily on current work, new applications, and emerging approaches for the mathematical characterization of essential aspects of molecular structure, while the other half describes applications of structural approach to new drug discovery, virtual screening, protein folding, predictive toxicology, DNA structure, and systems biology

Advances in molecular biology and toxicology are paving the way for major improvements in the evaluation of the hazards posed by the large number of chemicals found at low levels in the environment. The National Research Council was asked by the U.S. Environmental Protection Agency to review the state of the science and create a far-reaching vision for the future of toxicity testing. The book finds that developing, improving, and validating new laboratory tools based on recent scientific advances could significantly improve our ability to understand the hazards and risks posed by chemicals. This new knowledge would lead to much more informed environmental regulations and dramatically reduce the need for animal testing because the new tests would be based on human cells and cell components. Substantial scientific efforts and resources will be required to leverage these new technologies to realize the vision, but the result will be a more efficient, informative and less costly system for assessing the hazards posed by industrial chemicals and pesticides.
